Coral Gables, Florida 33134 Rabbit sperm fluoresced relatively uniformly following treatment with goat or rabbit antirabbit
semen globulin alone or in combination with heated complement. However, following an immobilizing treatment, namely, fluorescein conjugated goat or rabbit antirabbit semen globulin
plus active C', the sperm fluoresced minimally in the acrosomal region but brightly in the post
acrosomal area. This appearance corresponds to the position of the cell membrane of the sperm
head following the rolling back of the cell membrane seen by transmission electron microscopy
(Russo and Metz, 1974). Comparable posterior fluorescence was observed by indirect immunofluorescence. This posterior fluorescent pattern may be diagnostic for immunologically immobilized rabbit sperm. Membrane damage associated with immunological immobilization was also seen with SEM. This
appeared as an accumulation of material interpretable as a "rolling back" of the cell membrane
from the acrosomal apex.
