"Co-immunofluorescence of Zinc finger and BTB domain containing 16 (ZBTB16, previously known as PLZF) and amine oxidase (flavin containing) domain 2 (AOF2, previously known as LSD1) in isolated spermatogenic cells from adult mouse testes demonstrates stage-specific localization of AOF2. Early meiotic spermatocytes exhibit weak AOF2 expression (red), whereas highest levels of AOF2 protein can be detected in zygotene spermatocytes, postmeiotic round and elongating spermatids. The merged pictures (AOF2/ZBTB16) indicate a strong AOF2 signal (red) in ZBTB16 expressing spermatogonia (green). Cell nuclei were co-stained with DAPI (blue). ZBTB16 (green) is specifically expressed in type A spermatogonia. Roman numerals indicate the stage of the spermatogenic cycle. Elongating spermatids (ES), intermediate spermatogonia (In), leptotene spermatocytes (L), pachytene spermatocytes (P), round spermatids (RS), Sertoli cell (SC), spermatogonia type A (SGA; As = Asingle, Aal = Aaligned, A2), zygotene spermatocytes (Z)."

Immunohistochemical analyses of AOF2 in mouse testis sections at postnatal days (PND) 6, 10, 12, 14, 18 and 20 corresponding to the appearance of the spermatogenic cell types. Brown staining indicates positive reactivity. Roman numerals indicate the stage of the spermatogenic cycle. Round spermatids (RS), spermatogonia type B (B), spermatogonia type A (SGA), spermatogonia (SG), spermatocytes (Spc). For the negative control, the primary antibody was replaced by the corresponding concentration of rabbit IgG. Sections were counterstained with hematoxilin."
