High magnification confocal microscopy of epithelial sheets extracted from peri-ovulatory oviducts labeled with monkey sperm surface protein DEFB126 in situ demonstrates that DEFB126 localizes only to apical regions of secretory cells. The signal for DEFB126 (red) is superimposed on the differential interference contrast image. The labeling pattern is consistent with the distribution of sperm binding to oviductal epithelia in vitro as observed by both confocal and video micrography: 96% of the sperm attached to the epithelium bind (via the head) to secretory cells.
